ARCUT: Artificial Restriction DNA CUTter
A powerful tool for gene targeting of human genome

Abstract

We have been developing achemistry-based Artif icialRestriction DNA CUTter (ARCUT) composed of a pair ofpseudo-complementary peptide nucleic acids and CeIV/EDTAcomplex. This cutter has long (about 16 bp or more)recognition sequence, which enables us to cut huge DNAsuch as human genome at one target site. Here, we introducebriefly two of our latest studies. The first one demonstrates thatARCUT can cleave human genome at one target site. Thesecond one shows that ARCUT promotes the homologousrecombination repair in human cells. These results suggest thepossibility of gene targeting of human genome using ARCUT.
